Overview
DSIP (delta sleep-inducing peptide) is a nonapeptide first isolated in 1977 from the cerebral venous blood of rabbits undergoing induced slow-wave sleep. It carries the sequence Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu and is supplied as a lyophilised powder in a sealed vial for laboratory and in-vitro research.

Research Focus
DSIP takes its name from the delta-wave EEG activity observed after infusion in the original rabbit experiments. Early human work examined its influence on disturbed sleep, with published studies reporting shorter sleep onset latency and improved sleep efficiency, including effects measured on the following night rather than only immediately after administration.
Research interest has since extended well beyond sleep. Published work has examined DSIP in relation to stress-axis and corticotropin regulation, thermoregulation, analgesia, opioid withdrawal models and antioxidant activity — which is why the literature often describes it as multifunctional rather than sleep-specific.
It is worth being clear about the state of the evidence: despite the name, DSIP's status as an endogenous sleep factor remains unresolved. No corresponding gene, precursor protein or receptor has been identified, and findings across studies have been inconsistent — a 2006 review in the Journal of Neurochemistry characterised it as a still-unresolved riddle. DSIP remains an active subject of laboratory investigation rather than a settled one.
